- Use a suitable model to explain how separation and identification of a mixture of organic compounds can be achieved with a thin layer chromatographic (TLC) technique. What are some ways to visualize a TLC plate? Why may it be necessary to have more than one method to visualize a TLC plate?A student performs a TLC experiment and at the end of the experiment, sees no spots other than the initial spot. What does this mean, and what should the student do to fix the problem?
- Although TLC is qualitative and tells you what is present in a given sample, it can also be used to determine where a compound might be lost: What samples could you test on a single TLC plate that might give you this information?how does TLC determine if a sample is pure? Why chromatography is an important tool for analysis of organic compounds?
- 5. The following TLC plate was developed in 4:1 hexane/acetone. a) Draw what the TLC plate could look like if it were developed in 9:1 hexane/acetone. b) Draw what the TLC plate could look like if it were developed in 1:1 hexane/acetone. 4:1 hexane/acetone 9:1 hexane/acetone 1:1 hexane/acetoneHow does ammonia, dimethylglyoxime (DMG), and 8-hydroxyquinoline (8HQ) interact with your cations, leading to the evolution of colored species? Context: Paper chromatography experiment (separation of inorganic cations) with a 9:1 acetone/ HCl solvent1. In thin layer chromatography (TLC), explain how the pigments separate on a piece of paper. 2. If the spots are colorless on a TLC plate, how can you make it visible? Enumerate 2 ways and discuss each.
- How is TLC used to determine the purity of a sample?Question: Using a sample of aspirin for HPLC analysis. If the aspirin was contaminated (wet) with trace amounts of a solvent such as ethyl acetate, hexane, or water, would this contaminant be detected and appear as a peak in the HPLC chromatogram? Briefly explain based on the type of detector used in HPLC analysis and what type of compounds can be detected. My Answer: The HPLC analysis detects chromophores, compounds with multiple double bonds. Since water, ethyl acetate, and hexane all lack multiple pi bonds this will not be detected in the HPLC. -Question for you, is this complete (and correct), and should I include anything about UV detection? Is UV detection used in HPLC? I thought it was only used on TLC to show compunds that arent visable.TLC relies on differences in non-covalent forces. Explain the role of these non-covalent interactions in determining the Rf value obtained.
